Interaction study of mutated HAP40 and HTT. Human 293-based B1.21 cells, that express 17Q-HTT upon induction, were transfected with plasmids coding for either wtHAP40 or HAP40 mutants in which the E316, E317, E331, and D333 were replaced by lysine (HAP40-4 K). For the interaction study, wtHAP40 and HAP40 fused to a carboxy-terminal and amino-terminal TwinStrep-tag were used. Cell lysates (IP −) or elution (IP +) of the co-immunoprecipitation of wtHAP40 and HAP40-4 K using MagStrep beads (IBA) were analysed by Western-Blot.
